Mode of action
Work through three distinct mechanisms:
I. Alkyl groups are attached to DNA bases, causing repair enzymes to fragment the DNA to replace the alkylated bases, which prevents DNA synthesis and RNA transcription from the damaged DNA.
II. DNA damage via the formation of cross-links which prevents DNA from being separated for synthesis or transcription
III. the production of incorrect nucleotide pairing, which results in mutations.
Uses
Cancers of the ovaries, breast, blood and lymphatic systems, and nerves are all treated with cyclophosphamide (mainly in children).
