Mode of Action

  • PDE5 Inhibition: Tadalafil selectively inhibits PDE5, preventing the breakdown of cGMP.

  • cGMP Elevation: Leads to prolonged smooth muscle relaxation and vasodilation in the corpus cavernosum.

  • Sustained Erection: Facilitates erection by enhancing blood flow in response to sexual stimulation, with a longer duration of action compared to sildenafil.

Uses of Tadalafil

  • Erectile Dysfunction (ED): Treats erectile dysfunction by enhancing the natural erectile response.

  • Benign Prostatic Hyperplasia (BPH): Alleviates symptoms of BPH, such as difficulty in urination.

  • Pulmonary Arterial Hypertension (PAH): Used to lower pulmonary blood pressure, marketed as Adcirca®.
